大葱茎尖脱毒苗培养及增殖技术的研究

Studies on Culture and Propagation of Virus Elimination Seedings from Welsh Onion Shoot-tip

章丘大葱茎尖外植体启动培养基以MS +NAA 0 1mg·L-1 +2iP 0 4mg·L-1 最佳 ,诱导率为1 0 0 %,生根壮苗最佳培养基为 1 / 2MS +NAA 0 0 1mg·L-1 +IBA 1 5mg·L-1 ,生根率 1 0 0 %。驯化移栽成活率 1 0 0 %。分化培养基以MS +NAA 0 1mg·L-1 +2iP 0 6mg·L-1 最佳 ,丛生率 1 0 0 %,平均分化苗数6 8。根尖染色体观察 ,遗传表现稳定。

The results of virus elimination by shoot-tip culture of welsh onion commonly grown in Zhangqiu showed that the optimum medium for starting culture of shoot-tip was MS+NAA 0.1 mg·L -1+2iP 0.4 mg·L -1,the rate of shoot induction was 100%;the optimum medium for root induction was 1/2 MS+NAA 0.01 mg·L -1+IBA 1.5 mg·L -1,both rates of rooting were 100%,and survival rate after habituation were 100%。The optimum medium for shoot-tip to differentiate rank seedlings was MS+NAA 0.1 mg·L -1+2iP 0.6 mg·L -1,and the average divided seedings number of each shoot-tip was 6.8。The observation of cytology showed that this approach could maintain the stability of hereditary。
