摘要
采用改良CTAB法提取河南 17个桂花品种的基因组DNA ,利用RAPD技术对其进行鉴定和分类研究。从 10 0个 10bp的随机引物种筛选出 15个扩增效果较好的引物进行扩增 ,共产生 12 1条带 ,其中 87条为多态性带。根据扩增结果进行聚类分析 ,得出反映各品种间亲缘关系的树状图。各个品种群内的不同品种间的亲缘关系接近 ,而不同品种群间亲缘关系较远。RAPD对基因组的分析结果与传统分类学的结果基本相符。
Applying an improved CTAB method to extract DNA from leaves of Osmanthus fragrans, studied the classification and identification of 17 Osmanthus fragrans cultivars in Henan province by the technique of random amplified polymorphic DNA(RAPD). Fifteen better 10 bp primers which were selected from one hundred arbitrary primers were applied to be amplified. Total 121 bands were produced ,with which 87 bands were polymorphic. Genetic distance between each of the cultivars were calculated from simple matching coefficies. A dendrogram showing genetic relationships was constructed through an unweighted pair-group method. The genetic relationship of cultivar among different cultivar groups is farther than that in the same cultivar group.This conclusion is consistent with traditional cultivar classification. RAPD technology provides a few of new evidences for cultivar identification and classification to Osmanthus fragrans cultivars in Henan province.
