摘要
目的检测猪胃内是否存在幽门螺杆菌和其它螺杆菌。方法以一对幽门螺杆菌种特异性16SrDNA引物PCR扩增检测样品中幽门螺杆菌。合成三条螺杆菌属特异性引物,扩增螺杆菌16SrDNA部分序列检测螺杆菌,并通过限制性内切酶图谱和序列分析确定种。结果检测的48份样品中,幽门螺杆菌种特异性引物扩增全部呈阴性,但有50%样品存在螺杆菌。限制性酶切图谱和序列分析表明,存在三种螺杆菌,可能为Helicobactercanadensis、Helicobacterheilmannii和Helicobacterrodentium。结论所检测的猪胃标本未发现幽门螺杆菌,可能存在Helicobactercanadensis、Helicobacterheilmannii和Helicobacterrodentium。
To determine whether the helicobacter species exist in pig stomach, one pair of primers based on 16S rRNA specific to Helicobacter pylori (Hp)was used ro detect Hp in pig stomach.Three pieces primers specific to hilicobacter species were synthesized and used to amplify the partial sequences of 16S rRNA of the hilicobacter species,and then this species of bacteria in pig stomach were determined by restriction fragments length polymorphism (RFLP) and sequencing analysis.It was found that all the 48 samples showed negative results by amplifications with the Hp specific primers, but 50% of samples showed positive results in PCR RFLP analysis,suggesting the presence of helicobacter species, probably they might be H. canadensis, H.heilmannii and H.rodentium.
