肺炎球菌溶血素提取及其免疫学效果观察

Extraction and Immunological Effect of Pneumolysin

目的 提取 2 3F型肺炎球菌溶血素 (PLY) ,并对其免疫原性、抗原性及保护效果进行初步观察。方法 通过离子交换层析、疏水共价层析和排阻层析 ,从 2 3F型肺炎球菌中提取PLY ,以 2 0 μg的PLY免疫小鼠 (0、14及 2 1d) ,末次免疫后 14d用 2 3F、9V和 2型肺炎球菌从鼻腔粘膜攻击 ,观察保护效果。结果 成功获得PLY ,并能诱导小鼠产生较高水平的相应抗体 ;试验组小鼠存活率显著高于对照组小鼠。结论 PLY具有良好的免疫原性 ,对小鼠具有特异保护作用和交叉保护作用。

Objective To extract pneumolysin (PLY) of pneum oc occus of serotype 23F and study its immunogenicity,antigenicity and protective e ffect in mice.Methods Extract pneumolyin (PLY) from pneumococcu s of serotype 23F by ion exchange,hydrophobic covalent and exclusion chromatogra phy.Inject s.c. 3 doses of PLY into mice on days 0,14 and 21 respectively,20 μg per dose.Challenge the mice with S.pneumoniae strains of serotype 23F,9V an d 2 by nasal route 14 days after the 3rd injection and observe the protective effect of vaccine. Results PLY was successfully extracted,and high antibody level was induced in mice.The survival rate of mice in test group was significantly higher than that in control group. Conclusion The extracted PLY showed specific and cross protective eff ects on the mice challenged with pneumocci of different serotypes.
