茶多酚对高糖所致人肾小球系膜细胞Ⅳ型胶原合成的影响

摘要 目的 :探讨高糖对人肾小球系膜细胞Ⅳ型胶原合成的影响及茶多酚的干预作用。方法 :应用放射免疫法检测高糖条件下体外培养的系膜细胞上清液中Ⅳ型胶原含量及茶多酚干预后其含量的改变 ;应用免疫细胞化学法检测高糖条件下系膜细胞内Ⅳ型胶原的表达以及茶多酚干预后的变化。结果 :高糖可增加系膜细胞上清液及细胞内Ⅳ型胶原的含量 ;茶多酚可抑制高糖条件下上清液中及细胞内Ⅳ型胶原的增加。结论 :高糖可导致人肾小球系膜细胞Ⅳ型胶原合成增加 ,而茶多酚可有效干预此过程。 Objective To investigate the effect of high gluco se on the level of type Ⅳ collagen in human mesangial cells and the intervention of tea polypheno ls (TPs). Methods The cells were cultured in high glucose with or without TPs. The contents of type Ⅳ collagen in the media were detected by r adioimmunoassay. Immunocytochemistry was used to detect the expression of type Ⅳ collagen in the cells. Results Level of type Ⅳ collagen in human mesangial ce lls increased cul tured with high glucose,however,it decreased co-cultured with TPs compared with that only with high glucose. Conclusion High glucose contributes to an i ncrease of typ e Ⅳ collagen produced by human mesangial cells and TPs can reduce high glucose -induced type Ⅳ collagen.
