低氧条件下四逆汤及组方药提取物对大鼠血管内皮细胞释放PGI_2,TXA_2和NO的影响 被引量:9

摘要 目的 :观察四逆汤提取物 (DRE)及其组方药提取物对低氧条件下大鼠血管内皮细胞释放血栓素A2(TXA2 )、前列环素 (PGI2 )和一氧化氮 (NO)的影响。方法 :低氧处理的大鼠血管内皮细胞经四逆汤及其组方药提取物作用后 ,取条件培养液 ,采用放射免疫法及Griess反应测定培养液中的PGI2 ,TXA2 和NO的代谢产物 6 酮 前列腺素F1α(6 keto PGF1α) ,血栓素B2 (TXB2 )及亚硝酸盐 (NO2 -)含量。结果 :与对照组比较 ,DRE、甘草提取物 (LE)、干姜提取物 (GE)、附子提取物 (AE)、甘草加附子提取物 (ALE)、干姜加附子提取物 (AGE)可以显著增加低氧处理的大鼠血管内皮细胞培养液中 6 keto PGF1α的含量及 6 keto PGF1α/TXB2 的比值 ,但对TXB2 的含量没有显著影响。DRE组对 6 keto PGF1α含量的影响显著高于LE ,GE ,AE ,ALE ,AGE组。DRE组对 6 keto PGF1α/TXB2 比值的影响显著高于GE ,AE ,ALE组。与对照组比较 ,DRE ,LE ,GE ,AE ,AGE ,ALE组显著提高血管内皮细胞培养液中NO2 -的含量。DRE的药理作用显著高于GE ,AE ,ALE组。结论 :四逆汤提取物增加低氧处理的大鼠血管内皮细胞培养液中NO ,PGI2含量及PGI2 /TXA2 的比值。 Objective: To compare the effect of the extr ac ts from Decoction for resuscitation (DRE) and its component herbs on prostacycli n(PGI 2), thromboxane A 2(TXA 2) and nitric oxide (NO) release from rat va scular endothelial cells under hypoxia. Method: After treatmen t with the extracts from DRE and its component herbs, the contents of 6-keto- prostaglandin F1α(6-keto-PGF 1α ), thromboxane B 2 (TXB 2) as well as nitrite (NO), which were degradation products of PGI 2, TXA 2 and NO respec tively, in culture medium of rat vascular endothelial cells under hypoxia were measured with radioimmunoassay and Griess Reaction. Result: Co mpared with the control group, the results indicated that DRE, prepared licori ce root extract (LE), dried ginger extract (GE), aconite root extract (AE), e xtracts of aconite root and prepared licorice root (ALE), extracts of aconite r oot and dried ginger (AGE) increased significantly the content of 6-keto-PGF 1α and the ratio of 6-keto-PGF 1α /TXB 2, but had no effect on the content of TXB 2 in culture medium of rat vascular endothelial cells under hyp oxia. The content of 6-keto-PGF 1α in the DRE group was higher than tha t in the groups of LE, GE, AE, ALE, AGE. The ratio of 6-keto-PGF 1α /TXB 2 in the DRE group was higher than that of the groups of GE, AE, ALE . Compared with the control group, DRE, LE, GE, AE, ALE, AGE increased si gnificantly the content of NO 2 - in culture medium of rat vascular endotheli al cells under hypoxia. Moreover, the content of NO 2 - in the DRE group wa s higher than that of the groups of GE, AE, ALE. Conclusion: The results suggested that DRE increased significantly the content of PGI 2 an d the ratio of PGI 2/TXA 2 as well as the content of NO. The effect of DRE on the parameters in culture medium of rat vascular endothelial cells under hypoxi a was better than that of the extracts from its component herbs. [
