脑胶质瘤中iNOS、VEGF的表达与肿瘤血管形成的相关性和意义 被引量:1

RELATIONSHIP BETWEEN NITRIC OXIDE AND VASCULAR ENDOTHELIAL GROWTH FACTOR IN ANGIOGENESIS GLIOMA

摘要 目的探讨一氧化氮合酶 (induciblenitricoxidesynthase,iNOS)和血管内皮生长因子 (vascularendothelialgrowthfactor,VEGF)在脑胶质瘤血管形成中的作用。方法采用免疫组织化学法检测 5 0例不同级别胶质瘤标本中的诱导型iNOS、VEGF和Ⅷ因子相关抗原 (factorⅧrelatedantigen ,FⅧRAg)的表达 ,分析其相互关系。结果①对照组无iNOS表达 ,胶质瘤组iNOS阳性表达率为 4 0 .96 5± 316 ,二者有显著性差异 (P <0 .0 0 1) ;②iNOS阴性组微血管密度为 11.2 0 0± 3.4 91,iNOS阳性组微血管密度为 6 9.792± 5 18,二者间有显著性差异 (P <0 .0 5 ) ;③胶质瘤VEGF阳性表达为 75 .18± 6 .4 76 ,明显高于对照组 (P <0 .0 1) ;④iNOS阳性组中VEGF阳性细胞数明显高于对照组 (P <0 .0 1)。结论FⅧRag的检测能较好地反映脑胶质瘤血管生成活性 ;VEGF和iNOS可能通过相互上调表达促进血管生成 。 ObjectiveTo investigate tha roles of nitric oxide(NO) and vascular endothelial growth factor(VEGF) in glioma angiogenesis. MethodsThe expressions of inducible nitric oxide synthase(iNOS), VEGF and factor Ⅷ related antigen(FⅧRag) were evaluated by immunohistochemical technique, and their mutual relationships were analyzed in 50 cases of gliomas. Results①The positive expression rate of iNOS was 40.965±316 in glioma group, while no expression was found in control group, there was a significant difference between the two groups (P< 0.001). ②The microvascular density was 11.200± 3.491 in iNOS negative group and 69.792±518 in iNOS positive group, there was a significant difference between them (P< 0.05). ③The positive expression rate of VEGF was 75.18± 6.476 in glioma group, which was higher than that of the control group(P< 0.01). ④The VEGF positive cells in iNOS positive samples were more than those in iNOS negative ones(P< 0.01). ConclusionDetection of FⅧRag could reflect the angiogenesis activities in astrocytomas. VEGF and iNOS could up-regulate each other to promote angiogenesis and are of importance in determining the malignancey degree of astrocytomas.
