氦氖激光照射对兔实验性牙移动牙周组织VEGF表达的影响 被引量:4

摘要 目的 :探讨氦氖激光照射对兔实验性牙移动牙周组织血管内皮细胞生长因子 (VEGF)表达的影响。方法 :30只大耳白兔 ,随机分成 6组 ,即加力 1、 3、 5、 7、 14及 2 1d组 ,每组 5只。加力各组动物于双侧上颌第一磨牙至切牙间拴结不锈钢螺簧 ,0 .0 8N力拉上颌第一磨牙向近中移动。加力各组动物采用自身对照 ,右侧为氦氖激光照射侧 ,左侧为对照侧。用波长 6 32 .8nm、激光功率 2 0 m W的氦氖激光照射加力各组兔右侧颊部 ,除 1、3d组分别照射 1、 3次外 ,其余各组连续照射 5次 ,每天 1次。制作以上颌第一磨牙为中心的、近远中方向的组织切片 ,行 VEGF免疫组化染色。进行组织学观察及图像分析 ,对 VEGF免疫组化染色的灰度积分进行统计学处理。结果 :压力区加力 1d组照射侧 VEGF表达 (灰度值 ) (130 .0 2± 4 .2 9)明显低于对照侧 (136 .74± 4 .5 6 )(P<0 .0 5 ) ;而 3d组和 5 d组照射侧 (15 1.2 0± 2 .98和 16 2 .11± 4 .32 )明显高于对照侧 (14 2 .38± 4 .87和 15 1.10±6 .4 5 ) (P<0 .0 5 )。张力区加力 3、 5和 7d组照射侧的 VEGF表达 (灰度值 ) (14 3.96± 3.6 3、 14 8.4 0± 5 .79和15 0 .72± 5 .79)较对照侧 (138.2 0± 4 .88、 138.36± 6 .17和 14 0 .2 0± 6 .35 )明显升高 (P<0 .0 5 )。 Objective To investigate the effects of He-Ne laser irradiation on VEGF expression in periodontium of experimental tooth movement in rabbits.Methods Thirty rabbits were randomly divided into 6 groups on average: forced group of 1, 3, 5, 7, 14, 21 days. An orthodontic appliance, consisting of a coil spring ligated to the first molar connecting it to an orthodontic wire ligated onto the incisors, and exerting a force of approximately 0.08 N. The right side was designed as experimental side, left side as control side, in the self-control. Experimental side received irradiation of He-Ne laser. The animals from each group were killed at the time discontinued. The histological sections were stained with mouse anti-rabbit VEGF monoclonal antibody to express VEGF immunoreactivity, using a SP KIT, and counterstained with hematoxylin. Results The VEGF expression of irridiation side in forced group of 1 day (130.02±4.29) was significantly lower than that in control side (136.74±4.56)(P<0.05) in the pressure region,but the VEGF expressions of irridiation sides in forced groups of 3 days and 5 days (151.20±2.98 and 162.11±4.32) were significantly higher than those in control sides (142.38± 4.87 and 151.10±6.45)(P<0.05) in the pressure regions. The VEGF expressions of irridiation sides of forced group of 3,5, and 7 days (143.96±3.63,148.40±5.79, and 150.72±5.79) were remarkably increased, compared with control sides (138.20±4.88,138.36±6.17, and 140.20±6.35)(P<0.05) in the tension regions. Conclusion He-Ne laser irradiation can upregulate the expression of VEGF in periodontium of experimental tooth movement in rabbits.
