摘要
目的 探讨骨髓源干细胞迁移至损伤脑组织的可能性。 方法 致死剂量照射的C5 7BL 6雌性小鼠 ,接受月龄相配的雄性小鼠骨髓移植。骨髓移植 2个月后制作脑外伤模型 ,1、2、4和 6周分批处死存活的动物。分别在外周血涂片和新鲜脑切片上用荧光原位杂交法检测Y染色体阳性细胞。 结果 在移植后 6周的脑外伤病灶周围发现Y染色体阳性细胞。 结论 骨髓源干细胞可以迁移至脑外伤病灶周围 ,但这些细胞的性质需要进一步研究证实。
Objective To investigate whether bone marrowderived stem cells could migrate into the damaged brain parenchyma. Methods A cohort of lethally irradiated C57BL/6 female mice received whole bone marrow transplantation of age-matched male donors and survived from brain injury at 2 months from bone marrow transplantation, then were sacrificed at the 1,2,4 and 6 weeks after brain injury. Fluorescence in situ hybridization (FISH) for the Y chromosome was performed on the peripheral blood smear and fresh brain tissue sections. Results Y chromosome positive cells were found in the boundary area of damaged brain tissue at 6 weeks after brain injury. Conclusion These data suggest that bone marrowderived stem cells can entry into the boundary area of the damaged brain tissue. The further research is needed for identifying the characters of these cells.;
