摘要
目的 研究骨髓基质细胞(BMSc)的体外成骨能力,为骨组织工程研究提供细胞来源。方法 采用密度梯度离心法将兔BMSc自小量骨髓中分离培养,并观察第2代BMSc在地塞米松、维生素C、β-甘油磷酸钠诱导条件下的生长及成骨分化情况。结果 BMSc呈成纤维细胞样表现,增殖能力强,在诱导条件下碱性酸酶活性明显增高,并出现矿化结节。结论 BMSc体外增殖能力强,成骨能力肯定。地塞米松、维生素C、β-甘油磷酸钠可促进BMSc向成骨细胞分化。
Objective To study the osteogenic potential of cultured bone marrow stromal cells (BMSc) of rabbits in vitro and provide the suitable seed- cells for bone tissue engineering. Method BMSc of the rabbits were isolated by density gradient separation method and cultured, then BMSc at the second passage were cultured in subculture including dexamethasone, vitamin C and β-gly-cerophosphate, the proliferation capability and osteogenic differentiation were investigated. Results BMSc were spindle-shaped and have strong capability of proliferation. Alkaline phosphatase (ALP) activity of BMSc at third passage was increased in conditional medium and the mineral nodules were observed. Conclusion BMSc has a strong activity of proliferation and osteogenic potential, and dexamethasone, vitamin C and β-glycerophosphate condition the differentiation of BMSc into osteoblast-like cells.
