摘要
目的 通过体外实验观察生长激素(GH)对中性粒细胞凋亡和氧中间产物(ROI)的影响。方法 从健康儿童中取静脉血,全血洗涤后用生长激素(0或100 ng/ml)预处理,然后分别培养0、4和12 h,流式细胞仪测定亚二倍体、Fas表达和ROI的产生。结果 经过12 h培养,GH抑制中性粒细胞的凋亡,增加ROI的产生,同时降低Fas的表达。结论 尽管中性粒细胞的活化有其不利的一面,GH可以通过延长中性粒细胞的生命周期来改善机体的防御功能。
Objective To investigated the effects of PMN, reactive oxygen intermediates(ROI) production, and Fas expression. Methods Venous blood was collected from healthy children. Whole blood was washed and pretreated with GH for 3 h and then cultured for 0, 4, 12 h. Whole blood was analyzed by flow cytometry for cell death, Fas expression, and ROI. Results GH inhibited PMN apoptosis for 12 h of culture. GH enhanced ROI production by PMN throughout the 12 hour culture. GH decreased Fas expression on PMN for 4 h of culture. Conclusion Although activated PMN have potentially harmful aspects, GH may improve host defense.
