摘要
采用高效液相色谱法测定了长蛇Sauridaelongata新鲜肌肉中18种蛋白氨基酸的含量,结果表明,氨基酸总量为191 94mg/g,其中必需氨基酸(72 58mg/g)和鲜味氨基酸含量(91 59mg/g)皆较高,分别占氨基酸总量的37 81%和47 72%。该方法分离效果较好,重现性好,除半胱氨酸和组氨酸的相对标准偏差(RSD)为8 1%和8 8%外,其余氨基酸的RSD均在3 0%~6 5%之间;仅组氨酸的回收率较低,为72 0%,其余氨基酸的回收率皆为83%~111%,平均回收率为93 2%。该方法测定结果准确,其线性范围为10~1000μmol/L,检测极限为5μmol/L。
The contents of 18 kinds of amino acids in muscle of Saurida elongata were measured by high performance liquid chromatography (HPLC) with good separation and good precision. The result showed that the total contents of amino acids were 191.94 mg/g in muscles of Saurida elongata. The total contents of essential amino acids and tasty amino acids were 72.58 mg/g and 91.59 mg/g, accounting for 37.80% and 47.72% in total amino acids, respectively. There were relative standard deviations of 3.0%-6.5% in these amino acids, exception of Cys and His with 8.1% and 8.8%, respectively. The recoveries of these amino acids varied from 83% to 111% with an average of 93.2% (72% for His).
