摘要
目的 检测骨肉瘤细胞中凋亡抑制蛋白Survivin的表达及在胞内定位 ,研究其与骨肉瘤临床特征和血管内皮生长因子 (VEGF)表达的相关性 ,探讨Survivin在骨肉瘤发生发展及转移中的作用及机制。方法 采集骨肉瘤标本及其病例资料 ,采用免疫组织化学技术检测 4 6例骨肉瘤标本Survivin和VEGF的表达。统计分析Survivin与VEGF在骨肉瘤组织中表达的相关性和Survivin在骨肉瘤表达与病例临床特征之间的相关性。结果 4 6例骨肉瘤组织标本中2 6例Survivin蛋白表达阳性 (2 6 / 4 6 ) ,其中 2 1例胞质阳性 (2 1/ 4 6 ) ,4例胞核阳性 (4 / 4 6 ) ,1例胞质胞核均阳性(1/ 4 6 )。胞质Survivin阳性标本中 14例VEGF表达阳性 ,胞核Survivin阳性标本中 1例VEGF阳性表达 ,仅有 5例VEGF阳性而Survivin阴性。对照组骨纤维结构不良标本未检测到Survivin的表达。Survivin的表达与骨肉瘤的发病年龄、性别、肿瘤大小、原发部位、诊断时骨肉瘤有否转移均无关。结论 Survivin在骨肉瘤中的表达可能参与肿瘤的发生发展和转移 。
Objective To investigate the intracellular localization of Survivin protein in human osteosarcoma and its relationship with the clinical and pathological features and the expression vascular endothelial cell growth factor (VEGF) in osteosarcoma. Methods The expression of Suvivin and VEGF protein was detected by immunohistochemical assay in 46 cases of osteosarcoma. The relationship between the expression of Survivin and VEGF in osteosarcoma and the relationship between the expression of Survivin and clinical and pathological features were studied.Results In all the 46 cases, the Suvivin protein expression was detected in 26 cases. Among these positive cases, 21 cases (21/46) were positive in the cytoplasm, 4 cases (4/46) in the nucleus and one case in both cytoplasm and nucleus. Of the cases positive for cytoplasmic Survivin, 14 cases were positive for VEGF, and those positive for nuclear Survivin, one was positive for VEGF. There were also 5 cases of VEGF expression in Survivin negative samples. Neither the Suvivin nor the VEGF was detected in the cases of fibrous hyperplasia of bone. The Survivin expression was not correlated with the age, gender, tumor diameter, primary locations and metastasis at the time of diagnosis (P>0.05).Conclusion The expression of Suvivin may play an important role in the onset and progression of osteosarcoma. Survivin could take synergistic action with VEGF in the metastasis of osteosarcoma.
