摘要
目的 研究Rac 1蛋白和基质金属蛋白酶 2 (MMP 2 )在人卵巢癌中的表达 ,探讨其与卵巢癌组织学类型、临床分期、病理分化程度和转移的关系。方法 应用辣根过氧化物酶标记的链霉卵白素免疫组织化学方法 (SP法 )对 5 0例卵巢癌、 18例良性卵巢肿瘤及 12例正常卵巢组织中的Rac 1蛋白和MMP 2蛋白进行检测。结果 Rac 1蛋白和MMP 2蛋白在卵巢癌中的表达明显比正常卵巢组织及良性病变高 ,差异有显著性意义 (均P <0 0 1)。Rac 1和MMP 2在不同组织学类型标本中表达差异无统计学意义 (均P >0 0 5 )。Rac 1及MMP 2在不同临床分期、分化程度和肿瘤转移的标本中的阳性表达率比较 ,差异有显著性意义 (P <0 0 1或P <0 0 5 )。Rac 1和MMP 2两者在肿瘤转移过程中密切相关 (r =0 74 0 ,P <0 0 1)。结论 Rac 1和MMP 2蛋白的表达与卵巢癌的临床分期、分化程度及转移密切相关 ;Rac 1/MMP 2通路在卵巢癌的侵袭转移中起着重要作用。
Objective To study the expression of Rac-1 and matrix metalloproteinase-2 (MMP-2) in human ovarian carcinomas and to explore the correlation between the expression of Rac-1 and MMP-2 and the malignant status, including histological types, clinical stage, differentiation, metastasis.Methods Immunohistochemistry was used to measure the expression of Rac-1 and MMP-2 in 50 cases of human ovarian carcinomas, 18 cases of human benign ovarian tumors and 12 cases of human normal ovaries.Results The expression levels of Rac-1 and MMP-2 in the human ovarian carcinomas were higher than those in human benign ovarian tumors and normal ovaries with the difference being significant (P<0.01). No obvious difference in the expression of both Rac-1 and MMP-2 among the histological types of human ovarian carcinomas was found (P>0.05). There was significant difference in the expression of Rac-1 and MMP-2 among the clinical stages, differentiations and tumor metastases in human ovarian carcinomas (P<0.01 or P<0.05). The expression of Rac-1 was closely correlated with that of MMP-2 in the tumor metastasis (r=0.740, P<0.01).Conclusion The expression of Rac-1 and MMP-2 in human ovarian carcinomas is correlated with the clinical stage, differentiation and metastasis. Rac-1/MMP-2 signaling pathway plays a positive role in tumor cell invasion and metastasis.
