摘要
目的 研究Notch受 /配体在大鼠肺发育过程中的表达特征 ,探讨Notch信号对肺形态发生和肺泡上皮成熟分化的调控作用。方法 利用肺组织病理学检查、免疫组化、RT PCR半定量分析技术 ,检测Notch1~ 3 、Jagged1在大鼠肺发育不同阶段 (胎龄 18、 2 0、 2 1d及生后 1、 4、 7、 10、 14、 2 1d)肺组织内的表达。结果 大鼠肺发育囊泡期即有Notch受 /配体的显著表达 ,主要分布于气道上皮细胞、肺泡上皮细胞、血管内皮细胞和间质细胞 ,Notch1~ 3 及Jagged1蛋白及mRNA表达水平随胎龄增加呈动态改变。结论 Notch信号在肺发育中起重要作用 ,可能参与支气管树及气血屏障的形成 ,且对呼吸道上皮细胞、肺泡上皮细胞的成熟分化有重要调节作用。
Objective To investigate the temporal expression of Notch in lung development of rats and to explore the regulating role of Notch in lung development.Methods The expression of Notch 1—3 isforms and Jagged 1 was studied in the lungs of rats with gestation age of 18 days, 20 days, 21 days and postnatal 1 day, 4 days, 7 days, 10 days, 14 days, 21 days. Six rats of each group were used to assess lung histologic changes by means of histopathology and detect the expression of Notch in lungs by immunohistochemistry. Total RNA was extracted by Trizol reagent from the frozen lung tissues. mRNA levels of Notch was detected by reverse transcription polymerase chain reaction (RT-PCR). Results Notch 1—3 and Jagged 1 were mainly localized in the airway surface epithelium, alveolar epithelial cell during the pseudoglandular stage. The expression level of Notch 1—3 and Jagged 1 changed with the age and reached the peaks at canalicular period. Conclusion The presence of Notch signal was different at individual development stages and in different locations during the lung development respectively. These findings suggest that its functions are changing in different stages, and it may not only modulate the process of branching morphogenesis, but also may be involved in epithelial maturation and differentiation.
