摘要
目的 用高效液相色谱法 ( HPL C法 )测定炙甘草中甘草酸的含量 ,考察甘草炮制前后甘草酸的含量变化。方法 采用 C1 8柱 ,流动相为甲醇 - 0 .2 mol/L醋酸铵溶液 -冰醋酸 ( 6 7∶ 33∶ 1) ,流速为 1.0 ml/L ,检测波长为 2 5 0 nm。结果 甘草酸在 0 .80 16~ 2 .80 5 6μg范围内进样量与峰面积线性关系良好 ;平均加样回收率为 98.6 % ,RSD=1.4 0 % ( n=5 )。结论 HPL C法操作简便、易行 ,可满足定量分析标准要求 ;甘草经炮制加工成炙甘草 ,甘草酸含量降低 2 0
Object To investigate the content changes of Glycyrrhizic acid in Radix Glycyrrhizae before and after preparation, we used HPLC to determine the content of Glycyrrhizic acid in Radix Glycyrrhizae preparata.Method The separation was conducted on a Kromasil C 18 column with glacial acetic acid (67∶33∶1) as mobile phase, flow rate is 1.0 mL·min -1 ,detection wavelength at 250 nm.Results The linear relation of injection content with peak area was good in the ranges of 0.801 6~2.805 6μg;The average recovery was 98.6%. The RSD (n=5) was 1.40%.Conclusion This paper provides a rapid, convenient method which is suitable for quantitative analysis. The content of Glycyrrhizic acid is decreased by 20% after the Radix Glycyrrhizae is prepared.
