摘要
在红豆草单细胞悬浮培养过程中,我们观察到:培养初期大量细胞中胞质环流活跃,细胞内含物约在20min 内绕细胞环流一周;部分单细胞在悬浮培养早期就进行 DNA 复制;培养到72时,少数细胞开始分裂,这时还可见到无丝分裂;悬浮培养10~30d 中可观察到体细胞胚的发生和发育过程;40d 后达到肉眼可见的体细胞胚,转移到固体培养基后以很高的频率分化成苗.
The callus originated from the hypocotyl of Cnobrychis viciaefoliaScop.was suspended and shaked on a shaking machine.The isolatedsingle cells obtained from the shaking were then transferred to thinlayerLS liquid menium.During the later perio of suspension culture,it wasobserved that(1)the cyclosis was obvious and it took about 20 minutesfor one cirele;(2)some single cells were active in synthesis DNAduring the initial stage of suspension;(3)a few single cells startedto divide in about 72 hours and amitosis could be seen;(4)somaticembryogenesis could be also seen between 10-30 days;(5)a lot ofembryogenic cell masses were formed after 40 days of culture,and whentransferred to solid-medium they would differentiate into shoots at ahigh freqoency.
