摘要
目的探讨抗白介素10单克隆抗体在抗系统性白念珠菌感染中的作用。方法建立环磷酰胺诱导的免疫抑制小鼠系统性白念珠菌感染动物模型,设置对照组(单纯白念珠菌感染组)与处理组(白念珠菌感染同时注射抗白介素10单克隆抗体)。用平皿稀释法检测肾脏,脾脏组织菌落形成单位(cfu)数目;制作肝、肾、脾脏组织病理学标本,评估其病理学分级;并通过酶联免疫吸附试验(ELISA)检测脾脏干扰素γ分泌水平。结果感染后2d、3d、7d,处理组肾脏cfu值明显低于对照组(P<0.05);脾脏组织cfu值也低于对照组,但差异无显著性(P>0.05)。肾脏组织病理学评分处理组低于对照组,处理组较对照组感染程度减轻,差异有显著性(P<0.01);脾脏组织病理学评分处理组也低于对照组,但差异无显著性(P>0.05)。同时检测脾脏干扰素γ分泌水平处理组明显高于对照组,具有统计学意义。结论抗白介素10单克隆抗体在小鼠系统性白念珠菌感染中具有治疗作用。
Objective To investigate the effect of a monoclonal anti-interleukin-10 antibody (anti-IL-10 MAb) on systemic candidiasis in a murine model. Methods A cyclophosphamide-induced immunosuppressed murine model of systemic candidiasis was established. Two groups including the control group and the anti-IL-10 MAb treatment group were set in these mice. Colony forming units (cfu) of the kidney and spleen were determined with plating dilution method. The histopathological changes and degree of infection of the liver, spleen and kidney were graded. Meanwhile, the levels of interferon gamma (IFN-Results On the 2nd d, 3rd d and 7th d after C. albicans infection the numbers of CFU of the fungi in the kidney in the control group were significantly higher than those in the anti-IL-10 MAb treatment group (P < 0.05). The numbers of CFU of the fungi on the 2nd d, 3rd d and 7th d after C. albicans infection in the spleen in the control group were also higher than those in the treatment group but without statistically significant difference (P > 0.05). The scores of histopathological changes in the kidney in treatment group were lower than those in the control group (P < 0.01), and the degree of infection was milder in the treatment group. The scores of histopathological changes in the spleen in the treatment group were also lower than those in the control group but without statistically significant difference (P > 0.05). The levels splenic IFN-Conclusion It is demonstrated that the anti-IL-10 MAb has therapeutic effect on murine systemic candidiasis.
