诱变选育GOD酶活力高的黑曲霉菌株

摘要 以黑曲霉UCD69菌株为原始菌株,酶活为25.8μ/g,经过一次紫外诱变,筛选出UCD-69-4菌株,酶活为46.7μ/g,UCD-69-4菌株再经过一次γ-射线诱变,筛选出UC-19菌株,酶活为77.6μ/g. 通过实验,确定了几个影响菌体生长和产GOD的主要因素水平,其最适培养条件为旦白胨(动物)0.3%,蔗糖10%,MgSO_4·7H_2O0.01%,(NH_4)_2HPO_40.04%,在每个500ml三角瓶中装入100ml培养液,接种孢子量以200~300万个为宜,摇瓶培养32.5小时左右,温度30℃. A. niger UCD 69 strain was original strain, and the enzyme activity was 25.8μ/g. UCD-69-4 strain was selected from the original strain through mutagenesis (by ultraviolet rays), the enzyme activity was 46.7μ/g. Then the UC-19 strain was selected from UCD-69-4 strain by γ-rays, the enzyme activity was 77.6μ/g. Factors which effect on growth of the fungus and produce GOD were determined. The optimum conditions for culture ware peptong (animal) 0.3%, suger 10%, MgSO_4·7H_2O 0.01%,(NH_4)_2HPO_40.04%,100ml culture liquid in each flask of 500ml was sutable for inoculating spores 2000000-3000000, shaking flasks for 32.5 h at 30℃.
