摘要
目的 探讨人参皂甙 (GS)预处理给药及治疗性给药对离体大鼠心肌缺血再灌注损伤的保护作用的异同。方法 采用Langendorff离体心脏灌注模型 ,分对照组 (Con)、GS预处理组 (GSp)、GS治疗组 (GSt)。以Maclab 4S生理实验系统记录大鼠左心功能指标 ,包括左心室舒张压 (LVEDP)、左心室发展压 (LVDP)、左心室内压上升最大速率 (+dp dtmax)、左心室内压下降最大速率 (-dp dtmax)和心率 (HR)的波型和数值 ;用TTC染色法测定再灌注末左心室心肌梗死面积。结果 与对照组相比 ,GSp组和GSt组均显著提高心功能指数 ,减少心肌梗死面积 ;GSp组与GSt组间无差异。结论 GS预处理给药及治疗性给药对心肌缺血再灌注损伤均有明显的保护作用 ,二者心肌保护作用无显著差异。
Objective To investigate the protective effects of ginsenoside preconditioning (GSp) and ginsenoside treatment (GSt) on myocardial ischemia/reperfusion injury in isolated rat hearts. Methods 24 SD rats (weight 250~300 g) were randomly divided into 3 groups: control (ischemia-reperfusion group,Con), GSp and GSt. All of the hearts were perfused with Krebs-Henseleit (K-H) perfusate saturated with 95%O 2+5%CO 2 mixed gas. The ischemia reperfusion model was formed by inducing 30-min ischemia and 30-min reperfusion. The hearts in GSp group were given GS in K-H perfusate for 5 min followed by K-H for another 5 min prior to the induction of ischemia. To the hearts of GSt group, GS was given in the first 5 min of the 30-min reperfusion. LVEDP, LVDP, LVDP×HR and ±dp/dt max were recorded at the end of equilibration (perfusion with K-H for 30 min), 5 min before ischemia, and after 5, 10 and 20 min of reperfusion. Finally, the extent of myocardial infarct was measured. Results The cardiac function indexes in GSp and GSt groups were better than in the Con(P< 0.05,P< 0.01). The percentage of myocardial infract was less in GSp and GSt than in Con(P<0.05), but the difference in infarct between GDp and GSt was insignificant. Conclusion GS can protect isolated rat hearts against ischemia reperfusion injury, either for preconditioning or early treatment.
