肿瘤库(用于分子生物学研究)的建立及管理

Establishment and management of tumor bank applied for molecular biological researches

目的 探讨分子生物学研究中肿瘤库的建库条件 ,标本的采集、保存和质量控制 ,以及肿瘤库的信息化管理等有关问题。方法 手术或腔镜操作时采集的肿瘤组织 (包括肿瘤、瘤旁和远端正常组织 )以及肿瘤患者的血清 /血浆标本和从肿瘤组织中提取的核酸、从血液中分离的淋巴细胞、干细胞等肿瘤资源及正常人的血清标本按分子生物学实验要求分装处理后液氮或 80℃深低温冷冻保存 ;开发出一套信息管理系统应用于肿瘤库的管理 ,系统功能包括 :患者临床资料、随访信息、辅助检查信息、标本基本信息、标本存放位置坐标、标本使用与废除管理、实验结果登录、综合查询等。结果 1999年 12月至 2 0 0 3年12月收集标本达 3138份 ,包括良恶性肿瘤共 37个病种 /病理类型。已向市内外提供标本 32 0份 ,其中市内 2 4 0份 ,省外 80份 ,用于DNA、RNA和蛋白的相关研究 ,效果良好。结论 肿瘤库的建立 ,可向研究者们随时提供经确诊的、合乎现代分子肿瘤学研究要求的各种肿瘤标本和正常对照标本 。

Objective To explore the conditions of establishment of Wuxi city tumor bank applied for molecular biological researches, the methods of preparation and preservation of specimens, the quality control, and the management of tumor bank. Methods Tumor tissues including the tumor outsides and their normal counterparts, were derived from surgery or endoscopic procedures, serum/plasma specimen of tumor patients, nucleic acid extracted from tumor tissues, lymphocytes and stem cells isolated from blood and serum specimen of normal subjects. All were classified and processed according to requirements of molecular biological researches, then these specimen were stored in liquid nitrogen or at -80℃. A set of information management softwares were developed and applied to the management of clinical related studies of genomic DNA, RNA and protein. The results were satisfactory. Conclusion Various tumor specimens and their normal counterparts may be provided by tumor bank, which are pathologic confirmed and approaching the research requirements of modern molecular oncology. The establishment of tumor bank is helpful in sharing tumor information resources effectively.
