摘要
目的建立高低不同转移特性人成骨肉瘤MG63细胞亚系并研究其生物学特性。方法通过体外克隆技术和体内移植,筛选并建立了6个细胞亚系,测量其细胞电泳率、细胞增殖率,利用琼脂克隆形成实验和裸鼠体内成瘤实验,比较各细胞亚系的转移特性。结果A2、A3、A16亚系的细胞电泳率、细胞增殖率、琼脂克隆形成能力和肺转移灶形成率均明显高于A1、A6、A20亚系。结论通过体外克隆技术和体内移植的方法可建立不同转移特性的MG63细胞亚系,有助于骨肉瘤转移机理的进一步研究。
Objective:To establish human osteosarcoma MG 63 cell sublines with different metastatic capacities and study their biologic characters Method:Six cell sublines derived from human osteosarcoma MG 63 cell line were established by clone technique in vitro and transplantation in vivo The authors measured their cell electrophoretic and proliferatic rate and compared their metastatic capacities by using agarose clony formation and osteosarcoma formation in nude mice experiment Result:The cell electrophoretic and proliferatic rate,agarose clony formation ability and metastatic ability in lung of A2,A3 and A16 sublines were obviously higher than that of A1,A6 and A20 sublines Conclusion:The establishment of human osteosarcoma MG 63 cell sublines with different metastatic capacities is helpful for the further studies of metastatic mechanism
