摘要
籽粒硬度是决定小麦品质优劣的重要性状,控制硬度的主效基因为pinA和pinB(统称为puroindoline基因)。本研究利用含有能促进外源基因高效表达的赘序列、Kozak序列和poly(A)序列的pG4AB及含有Ubi启动子和筛选基因bar的pAHC25,成功构建了含有puroindoline基因的单子叶植物高效表达载体pUBPa和pUBPb。目的是将来通过转基因植物研究,进一步明确pinA和pinB的功能, 为通过基因工程方法快速改良我国现有小麦优良品种的籽粒硬度奠定基础。
Grain hardness is an important factor in determining wheat end-use quality. The major gene controlling grain hardness was pinA and pinB(puroindolines genes). In this study, high efficient expression vectors harboring wheat puroindoline genes were constructed using pG4AB and pAHC25, which harbor the ?赘 sequence, Kozak sequence and poly(A) sequence essential for higher level expression of foreign genes in transgenic plants, and the ubiquitin promoter and selection gene bar, respectively, as basic plasmids. The objective of this study is to further understand the function of puroindoline genes through transgenic plants and provide information for improving grain hardness through genetic engineering.
