铜绿假单胞菌主动外排系统MexAB-OprM与耐药的关系研究 被引量:30

摘要 目的 研究铜绿假单胞菌对碳青酶烯类抗生素的耐药机制。方法 以亚胺培南为代表 ,应用逆转录 (RT) PCR方法并设置内参照 ,分别研究临床分离的铜绿假单胞菌亚胺培南耐药株和亚胺培南敏感株的主动外排系统MexAB OprM中OprM的结构基因OprM的mRNA表达水平。结果 耐药组OprM的mRNA表达水平明显高于敏感组。结论 主动外排系统MexAB OprM的表达水平增高或异常增多与铜绿假单胞菌对以亚胺培南为代表的碳青酶烯类抗生素耐药密切相关。 OBJECTIVE To study the mechanism of imipenem resistance in clinical isolates of Pseudomonas aeruginosa. METHODS OprM gene is one part of the active efflux pump MexAB-OprM gene, the level of mRNA expressions of OprM in clinical P. aeruginosa isolates was determined by RT-PCR, and rpoD gene as an intrinsic control. RESULTS The levels of mRNA expression of OprM in imipenem-resistant strains were higher than those in susceptible ones(P<0.05). CONCLUSIONS The study indicated that the mRNA level of expressions of MexAB-OprM determines the level of imipenem-resistance in P. aeruginosa.
