反相高效液相色谱法测定大黄柳叶中地奥辛含量

摘要  目的:建立高效液相色谱法测定大黄柳叶中地奥辛含量。方法:分析柱为Zorbax SB C18(4.6 mm×150 mm,5μm),流动相为甲醇-水(40:60),流速1.0 mL·min-1,检测波长254 nm,柱温25℃,进样量20μL,用外标法定量。结果:线性范围在3.16-20.16μg内,r=0.9996,精密度实验RSD为1.5%,稳定性实验RSD为2.0%,平均回收率为97.5%,RSD为1.6%(n=5)。结论:方法简便、快速、有效、准确,重现性好,可用于大黄柳叶中黄酮类成分的定量分析。 Objective: HPLC method was established for determination of raddeanalin [ doismetin 7 -O-α-L-ar-abinopynosyl (1→6) - β- D - glucopyranoside ] in Salix raddeana Laksch samples of different areas and collected dates. Method:The analytical column was Zorbax SB C18(4. 6 mm×150 mm, 5 μm) ,the chromatography was carried out with a linear gradient program. The mobile phase consisted of MeOH - H2O(40: 60), at flow rate of 1.0 mL· min-1, peaks were detected at 254 nm, column temperature was at 25℃. Results:The linear range was 3. 160 -20.16μg(r =0. 9996). The average recovery was 97. 5% ,RSD 1. 6% (n =5). Conclusion:The method is accurate , simple, effective and feasible. It can be used to determine flavones in Salix raddeana Laksch.
