摘要
目的 通过观察 p16、p2 1WAF1、Rb、cyclinE 4种细胞周期相关蛋白在宫颈鳞状细胞癌中的表达 ,探讨它们在宫颈癌的细胞周期调控中的作用。方法 采用免疫组化EliVision二步法对 88例宫颈鳞癌组织 ,16例宫颈上皮内病变 (CIN)组织 ,15例宫颈炎组织进行 p16、p2 1WAF1、Rb、cyclinE 4种蛋白表达的检测。 结果 p16、p2 1WAF1、cyclinE在宫颈癌中的表达高于宫颈炎(P <0 0 5 ) ;Rb在宫颈癌的表达少于宫颈炎 (P <0 0 5 ) ;Rb与p16在宫颈癌中的表达呈负相关 (r =- 0 6 75 ,P <0 0 5 )。结论 宫颈癌细胞周期G1期中 ,由于Rb蛋白的缺失 ,使cyclinE表达升高 ,致使癌细胞增生 ;同时 ,p16、p2 1WAF1蛋白在宫颈癌中的表达增高 ,并失去抑制作用。
Purpose To investigate the expression of 4 G 1-S related proteins, p16, p21 WAF1 , Rb, in human cervical squamous carcinoma, and to discuss their roles in G 1-S cell cycle regulation. Methods Four proteins expression was observed immunohistochemically in 88 cervical squamous carcinomas, 16 cervical intraepithelial neoplasia (CIN) and 15 cervicitises respectively. Results The expression of p16, p21 WAF1 , and cyclinE in cervical carcinomas were higher than in cervicitises (P<0.05). The expression of Rb in cervical carcinomas was lower than in CINs and in cervicitises (P<0.05). There was a significantly negative correlation between the expression of Rb and p16 in cervical carcinomas (r=-0.675, P<0.05).Conclusions Rb protein is deleted cervical carcinoma, which makes higher cyclin E expression and unlimited proliferation of tumor cells; the repression functions of p16 and p21 WAF1 do not show in cervical carcinoma because of the overexpression.
