逆转录-实时定量PCR检测胃癌和肠癌病人CEA基因的表达 被引量:6

摘要 目的:通过检测胃癌和肠癌病人外周血中肿瘤细胞的CEAmRNA拷贝数,以实现对其肿瘤血运转移的定量监测。方法:采用逆转录-实时定量PCR技术检测65例正常人外周血和48例胃癌和肠癌病人外周血及肿瘤组织中CEAmRNA的拷贝数。结果:65例正常人外周血中无一检测到CEA基因转录本。经组织病理学诊断为恶性肿瘤的组织中CEA表达率为100%,CEAmRNA拷贝数在105~107/mg。48例肿瘤病人外周血CEA表达率为33.3%,CEAmRNA拷贝数范围在(101~103.2)/ml。结论:逆转录-实时定量PCR技术能特异、灵敏地检测胃癌和肠癌病人外周血中CEAmRNA,能早期客观地反映病人体内微转移的状况。 Objective: To establish a method for quantitative detection of CEA gene expression in the peripheral blood of patients with gastric or intestinal cancer. Methods: Real-time RT-PCR was taken to detect the CEAmRNA copy number in cancer tissues, as well as in the peripheral blood of 65 normal individuals and 48 patients. Results: CEA mRNA was totally expressed in cancer tissues of all 48 patients(100%), and the copy number was 105-107 per milligram. Also it was expressed thirty-three percent in peripheral blood in 16 out of 48 patients. Meanwhile, no CEAmRNA was detected in 65 normal individuals. Conclusions: Quantitative detection of CEA gene expression in the peripheral blood by the method of real-time RT-PCR is feasible for evaluating metastastic status of gastric or intestinal cancer.
