摘要 本文介绍速率法测定血清甘氨酰脑氨酸二肽氨基肽酶(Glycylprolinc dipcptidylaminopeptidase,GPDA)的方法.并应用于自动生化分析仪,测定的最适pH 为8.6,酶活力在1300 U/L 以下成线性,三份高、中、低值酶活力标本批内CV 0.7%~1.4%,批间CV1.2%~4.4%.132例正常人GPDA 为185.2±28.6((?)±s)U/L,65例原发性肝癌组为408.0±216.1 U/L((?)±s),明显高于正常组(p<0.001),且与AFP 水平无相关意义(r=0.0361),24例胃癌和18例肺癌,分别为99.5±16.6 U/L 和111.8±23.0U/L 明显低于正常组(p<0.001),并发现GPDA 在食道癌及贲门癌病人亦有下降趋势(p<0.01)。 A kinetic assay of serum glycylproline dipeptidyl aminopeptidasc(GPDA)was rcported in thispaper,it was used successfully for automatic analyzcr.Optimum PH was at 8.6.The enzyme activity was linear be-low 1300 U/L.The preeision of the method was examined by using three samples of sera with high,intermediateand low enzyme activity respectively.Within and between-assay CV ranged from 0.7 to 1.4% and from 1.2 to4.4% respectively.Serum GPDA in 132 healthy subjects was 185.2±28.6(x±s)u/L.The GPDA activily in serafrom 65 patients with primary hepatic cancer was 408.0±216.1u/L(x±s)and this value was significantly high.er than in healthy controls(P<0.001).No Correlation with alpha-fetoprotein was found(r=0.0361).TheGPDA in 24 patients with gastric cancer and 18 patients with lung careinoma was 99.5±16.6u/L and 111.8±23.0u/L.respectively,and these value were significantly lower than in healthy controls(P<0.001).The GPDAactivity in sera was found to be decreased in patients with esophagus and cardiac cancer(P<0.01).
