摘要
本文报告普鲁卡因、加热、普鲁卡因联合加热对15例急性非淋巴细胞白血病患者骨髓的白血病祖细胞和正常骨髓粒单祖细胞的作用。普鲁卡因联合加热可以显著增强对白血病祖细胞的抑制作用,对正常人粒单祖细胞影响较小。单独应用42℃水浴加热1小时后作细胞培养,白血病祖细胞集落形成单位残存1/5左右;用2mM普鲁卡因则其残存不到1/20。后者明显提高了对白血病祖细胞的体外杀伤效果。而后,我们对1例急性早幼粒细胞白血病患者的骨髓用后一方法体外净化并进行移植,患者骨髓回输无任何不适反应,造血功能恢复良好。
Procaine and hyperthermia have been shown to possess a relativelyselective cytotoxicity to leukemic cells. In this study. the combined effects of procaineand hyperthermia on the growth of hematopoietic progenitors (GM-CFU) andleukemic progenitors (L-CFU) were examined to determine if this combination result- ed in a great selective killing of leukemic cells than that achieved by procaine or heatalone. When the cells were treated simultaneously with procaine (2mM) and hyper-thermia(42℃) for one hour, the killing of L-CFU was enhanced considerably whereasGM-CFU were not markedly affected. These data indicate that the combined treat-ment with procaine and hyperthermia might offer an efficient mean to selectivelypurge residual leukemic cells in vitro. Procaine with hyperthermia may have a role inclinical autologous bone marrow transplantation for acute non-lymphocytic leukemia.
