摘要
以微胶囊法固定化磷脂酶A1,考察固定化方法、固定化载体和固定化条件与固定化酶的活力回收的关系,结果表明,微胶囊包埋法固定化磷脂酶A1的最佳条件是:壳聚糖(DD%=85%、分子量为1.3×105)浓度为1.5%、海藻酸钠浓度为3%、戊二醛浓度1%、钙离子浓度为0.5mol/L、pH值为6。固定化酶的活力回收达到81%,半衰期为350h。
PLA1(phospholipase A1) was immobilizied by the methods of microcapsule. There studied the meathods of immobilization,immobilized carriers and dependence between recovery of enzyme activity and conditions of immobilizied PLA1. The results showed that the condition of microcapsule for immobilizied PLA1 were the chitosan(DD%=85%,Mw=1.3×105)concentration 1.5%,alginate concentration 3%,glutaradehyde concentration 1%,Ca2+ concentration 0.5moL/L,pH=6,Finally getting recovery of enzyme activity 81% and T1/2=350h.
