亚低温对急性脑梗死大鼠脑组织Ca^(2+)和Mg^(2+)及兴奋性氨基酸与血浆内皮素变化的影响(英文) 被引量:2

摘要 背景:急性脑梗死后脑组织内钙离子(Ca2+)、兴奋性氨基酸(excitatoryaminoacids,EAA)和血浆内皮素含量的升高及脑组织内镁离子(Mg2+)含量的降低都会加重脑组织的损害。给予亚低温干预后,观察急性脑梗死大鼠脑组织Ca2+和Mg2+及EAA及血浆内皮素的变化,了解亚低温的脑保护作用。目的:探讨亚低温对大鼠急性脑梗死后脑组织内Ca2+及Mg2+和EAA、血浆内皮素变化的影响及意义。设计:随机对照实验研究。单位:武汉大学人民医院神经科。材料:实验于2000-01/2000-12在武汉大学人民医院神经科实验室完成。将48只SD大鼠随机分为亚低温组及对照组,各组再分为4个亚组,每亚组6只。干预:应用改良线拴法制备大鼠大脑中动脉梗死模型;亚低温组大鼠给予亚低温治疗,而对照组不作亚低温处理。分别于缺血后1,2,4,8h处死。主要观察指标:检测各个时段大鼠缺血区脑组织内Ca2+,Mg2+,EAA及血浆内皮素含量。结果:亚低温组大鼠缺血区脑组织内Ca2+,Mg2+,EAA及血浆内皮素含量随着缺血时间的延长仅轻度升高,Mg2+含量的下降也不明显,与对照组同时段比较有显著性差异(t=62.36~135.63,P<0.01)。结论:亚低温能明显阻止实验动物脑缺血后缺血区脑组织内损伤性因子Ca2+,EAA和血浆内皮素含量的增高及保护性因子Mg2+的下降,具有脑保护作用。 BACKGROUND:The increased content of Ca2+,excitatory amino acids(EAA) and endothelin(ET) as well as the decreased Mg2+in brain tissue with acute cerebral infarction will aggravate the damage of brain.To observe the changes of Ca2+,Mg2+and ET in brain tissues of mice after acute cerebral infarction happened in order to know the protective effects of subhypothermia after intervention of subhypothermia. OBJECTIVES:To explore the effects and significance of subhypothermia on Ca2+,Mg2+and EAA in brain of mice suffered from acute cerebral infarction. DESIGN:A randomized case control study. SETTING:Department of Neurology,People's Hospital of Wuhan University. MATERIALS:The experiment was completed in the Neurological Laboratory of People's Hospital in Wuhan Univeristy from January to December in 2000.A total of 48 SD mice were randomly divided into subhypothermia group and control group,each was divided into four subgroups with 6 mice in each. INTERVENTIONS:Improved thread embolism method was used to make middle cerebral artery infracted model.Subhypothermia treatment was conducted to subhypothermia group while no treatment for control group.Mice were executed in the 1st, 2nd, 4th and 8th hour after infarction. MAIN OUTCOME MEASURES:The content of Ca2+, Mg2+, EAA and ET in ischemic brain tissue in different time period. RESULTS:The content of Ca2+,Mg2+,EAA and ET in ischemia brain tissues of mice with subhypothermia intervention slightly increased with the advance of ischemic time and there was merely change of Mg2+.Compared with control group,there was significant difference(t=62.36-135.63,P< 0.01). CONCLUSION:Subhypothermia method can greatly inhibit the increase of damage factors such as Ca2+,EAA and ET in ischemic brain tissues of experimental animals and the decrease of protective factor Mg2+.
