摘要
背景:胆碱能系统与行为学研究相结合,已被广泛应用于包括中医药在内的益智和治疗老年性痴呆的研究。目的:探讨针药结合改善大鼠学习记忆能力的中枢胆碱能机制。设计:随机对照的试验研究。单位:江苏省中医院检验科,南京中医药大学第二临床医学院。材料:实验于2000-10/2001-02在南京中医药大学实验动物中心完成,50只健康9个月龄SD雄性大鼠,体质量(300±10)g。方法:将大鼠随机分为5组:空白组、模型组、尼莫地平组、针刺组及针药组。测定鼠大脑皮质乙酰胆碱酯酶活性(AchE)和乙酰胆碱(Ach)含量及跳台回避能力。主要观察指标:①跳台测试结果。②大鼠皮质的AchE活性和Ach含量。结果:东莨菪碱造模大鼠中枢胆碱能功能降低,学习、记忆错误次数增加,学习和测试错误次数空白组为1.70±0.34,0.50±0.17,模型组为3.90±0.48,2.20±0.70。针刺、针药结合可拮抗东莨菪碱诱发的学习记忆障碍(错误次数分别为2.00±0.21,0.80±0.25;1.80±0.63,0.60±0.84),提高乙酰胆碱的含量,降低AchE和Ach两者在脑组织中的比例(AchE/Ach),且针药结合作用更优。结论:针药结合改善学习记忆能力的机制可能与中枢胆碱能有关。
BACKGROUND:The collaboration of cholinergic system and behavior science has been widely applied in the researches,including in Chinese medicine, on benefiting intelligence and treatment of senile dementia. OBJECTIVE:To probe into the central cholinergic mechanism of the combination of acupuncture and medication on improvement of learning and memory abilities in rats. DESIGN:Aandomized and controlled trial study. SETTING:The research was performed in Examination Department of Jiangsu University of Chinese Medicine and the Second Clinical Medical Hospital Affiliated of Nanjing University of Chinese Medicine. MATERIALS:The experiment was performed in Experimental Animal Center of Nanjing University of Chinese Medicine from October 2000 to February 2001.A total of 50 normal SD male rats were employed, aged 9 months, weighted(300±10) g. METHODS:Rats were randomized into 5 groups:blank group,model group,nimodipine group, acupuncture group, acupuncture and medication group(A and M group).The activity of cerebral cortical acetylcholine esterase(AchE) and acetylcholine(Ach) amount as well as the ability of jump platform and escaping of rats were tested. MAIN OUTCOME MEASURES:The result of jump platform,AchE activity and Ach amount of cerebral cortex of rats. RESULTS:In scopolamine model rats,the central cholinergic function was declined and more mistakes of learning and memory were made.The frequency of learning and testing mistakes were(1.70±0.34),(0.50±0.17) in blank group,were(3.90±0.48),(2.20±0.70) in model group. Acupuncture and the combination of acupuncture and medication can be against learning and memory disorders induced by scopolamine[the frequency of mistakes were (2.00±0.21),(0.80±0.25);(1.80±0.63),(0.60 ±0.84) respectively],increase Ach amount, reduce the ratio of AchE and Ach in the brain tissues(AchE/Ach),and the results achieved by the combination of acupuncture and medication were much better. CONCLUSION:Mechanism of combination of acupuncture and medication on improvement of learning and memory abilities is probably related to central cholinergic system.
