nm23-h1基因转染对腺样囊性癌ACC-M细胞系淋巴结转移的影响

The Influence of Nm23-h1 Transfection on the Lymphatic Metastasis of Adnoid Cystic Carcinoma(ACC-M)Cell Lines

摘要 目的 :研究阳离子脂质体介导nm2 3-h1质粒转染腺样囊性癌ACC -M细胞株对其颌面部移植瘤区域淋巴结转移的影响。材料与方法 :体外用阳离子脂质体 (Lipofectamine)介导将nm2 3-h1质粒转染nm2 3-低表达的腺样囊性癌ACC -M高转移细胞株 ,经G4 18筛选得到高表达nm2 3-h1的细胞并以 5× 10 5/只植入 10只裸鼠颌下区 ,以未转染的细胞作对照。观察肿瘤生长及淋巴结转移情况。 4周后处死动物 ,获取标本 ,作组织病理学检查。结果 :未转染组有一只裸鼠颌下淋巴结转移 (1/ 10 ) ,4只 (4/ 10 )裸鼠淋巴结反应性增生 ,转染组未见淋巴结转移及明显增生。结论 :nm2 3-h1可能在腺样囊性癌的区域淋巴结转移中起重要的作用。 Objective:To study the influence of cationic lipid(Lipofectamine)-Mediated nm23-h1 transfection on the local lymphatic metastasis of salivary adnoid cystic carcinoma.Materials and methods:After in vitro incubation and transfection of nm23-h1 into the cells of low level nm23-h1-expressing ACC-M cell lines,the positively expressing cells were gained and transplanted to the submandibular region of 10 BALB/C mice,5×10 5 for each mouse and meanwhile,nontransfected cells was taken as control.Four weeks later,the mice were killed and the specimen were fixed in formalin and then pathological study was performed.Results:One mouse(1/10)in the nontransfected group had submandibular lymphatic metastasis,4 mice(4/10)in the same group has reactive lymphatic proliferation.There was no obvious lymphatic proliferation or metastasis in the transfected group.Conclusion:nm23-h1 might play a role in the local lymphatic metastasis process of adnoid cystic carcinoma.
