摘要
目的:探讨体外培养源于SD大鼠磨牙牙骨质的类成牙骨质细胞的可行性。方法:仔细取得12周龄SD大鼠第一磨牙,使用组织块结合酶消化的二步法体外培养源于牙骨质的类成牙骨质细胞,进行形态观察,测定其生长曲线、贴壁率,并以牙周韧带成纤维细胞为对照进行骨钙素免疫组化染色测定。结果:SD大鼠的类成牙骨质细胞约在第14~18天从根面长出;生长曲线呈“S”形;接种后12小时约有94%的细胞贴壁。骨钙素在体外培养的类成牙骨质细胞中被强烈表达,在成纤维细胞中几乎不表达。结论:SD大鼠的类成牙骨质细胞可以在体外成功培养。
Objective: To investigate the feasibility of culturing the SD rats cementoblasts-like in vitro. Methods: Themolar roots were obtained from 12-week-old SD rats. Using a 2-step method of explant culture combined enzyme digestion,cementoblasts-like were obtained from root surface of SD rat first molars and cultured in vitro.The shape of cementoblast-like were observed . Its growth curve and adherence rate were mensurated compared with the fibroblasts? Results: Thecementoblasts-like grew from the root surface between days 14 and 18. The growth curve showed. an ?S?shape . Theadherence rate after inoculation 12 hours is 94%.The cementoblasts-like manifested a polygonal shape and had twomorphotypes:osteoblasts-like and cuboidal or stratified.OCN was localized and manifested with a higher level expressionin cementoblasts-like in vitro . Fibroblasts lacked OCN activity as a control group. Conclusion: The cells consistent witha cementoblast-like phenotype of SD rat may be successfully cultured.
