抗人A型红细胞单克隆抗体的研究及其初步应用

摘要 在用人A型红细胞悬液免疫的Balb/c小鼠脾细胞与同系小鼠的骨髓瘤细胞进行融合屡遭失败的情况下,改用实体瘤分离出的骨髓瘤细胞进行融合,终获成功。融合后筛选出分泌盐水凝集抗体的杂交瘤细胞株,即为:3B_2、3H_2、8C_3细胞株。3株抗人A型红细胞单克隆抗体3B_2、3H_2、8C_3血清学检验和应用结果表明:3株单抗均可作为血型诊断试剂,可用以取代人源标准血清。 The titre of McAb 3B2, 3H2, 8C3 in culture and ascites are above 1 : 32 and 1 : 6400 respectively. Their affinities are 10~25 seconds and 4~6 seconds separately Through agglutination-inhibition test and absorptionelution test, we found that the McAbs were inhibited by type A blood group saliva and were first absorbed and the eluted by type A erythrocytes. Cold agglutinin has not been found in the McAbs. In the hemagglutination test ( 37℃ and room temperature) ,papain-test, anti-globulin test, the McAbs have not reacted with standard erythrocyte groups expressing 28 antigens of 10 blood group systems. While the 3 McAbs agglutinated both fetal and adult red blood cells, they did not reacted with red blood cells of monkey, dog, cow, rabbit, pig, fish, sheep, duck, chicken, mouse, pigeon and guinea pig. Thus, the 3 McAbs have reached the demand of blood grouping reagent and serve as useful substitutes for commercial standard anti-A grouping reagent derived from human source.
