摘要
经大、小鼠后海穴注射抗原后,应用免疫组织化学方法观察中枢神经系统内c-fos基因在不同时程的表达情况。结果发现与穴位神经分布相关的脊髓节段及下丘脑的一些核团和区域的c-fos表达高峰在2d和4d,其Fos免疫反应阳性(Foslikeimmunoreactivity,Fos-LI)神经元的数目及分布区域随时间的变化而不同。骶髓的Fos-LI神经元在2d组分布于Ⅱ→Ⅳ板层,4d组分布到Ⅳ→Ⅶ和Ⅹ板层;下丘脑的视前内侧区、结节区、下丘脑前核群、下丘脑腹内侧核和背内侧核等处的Fos-LI神经元数目在2d组和4d组分布有所不同。据实验结果推导:穴位免疫注射后,免疫信息经免疫体液因子在激活下丘脑神经内分泌免疫系统的同时;也激活与穴位相关的脊髓节段神经元的活动,经神经通路直接将周围的免疫信息传导到高位中枢,多途径、双重效应地激活免疫-神经内分泌网络,从而起到增强免疫效果的终效应。
Fos protein was examined immunohistochemically in the central nervous system at certain time points after bovine serum albumin as immunogen was injected into Houhai acupoint in rats and mice.The maximal peak of the appearance of Fos-like immunoreactivity(FosLI)neurons was observed on the second day and the fourth day in spinal segments relating to nerve distribution of the acupoint and in certain nuclei and areas in the hypothalamus.Numbers and areas of Fos-LI neurons varied from day to day. Fos-LI neurons in the sacral spinal cord distributed in Ⅱ→Ⅳ lamina on the second day,and in Ⅳ→Ⅶ andⅩlamina on the fourth day.Numbers of Fos-LI neurons were also different in the medial preoptic area and nuclei,the tuber cinereum area,the anterior hypothalamic nuclei,the ventromedial hypoth lamic nucleus and dorsomedial nucleus on the second and the fourth day.Our experimentsuggested that immuno-injection of Houhai acupoint may directly effect the neuroendocrine and neuroimmune systems of the hypothalamus by the way of immuno-fluid factors and may also effect the spinal neurouns in the segments relating to acupoint at the same time.Immuno-messages are transmitted from periphery to the hypothalamus directly,or from spinal neurons relating Houhai to the hypothalamus indirectly,and both direct and indirect neuroimmune pathways might double the stimulation on the neuro-immune network shown byFos-LI neurons in the present experiment.
