摘要
培养人脐带静脉内皮细胞(EC),提取人血浆低密度脂蛋白(LDL)并制成氧化型低密度脂蛋白(oxLDL),分别与血管EC培养后提取各组RNA,进行电泳及Northern印迹杂交,用人β-actin基因探针和人PECAM基因探针与各组RNA进行杂交。结果显示:EC在E-GMUV培养液中生长度好并可多次传代培养;用oxLDL处理EC24h可导致细胞死亡;用一步法提取单层培养EC的RNA效果较好;用地高辛标记人β-actin基因探针与各组RNA杂交均可见杂交带,而未见PECAM基因表达。提示单层培养人血管FC适于基因表达的研究;oxLDL对血管EC有细胞毒性,但不能促进PECAM基因表达。

low density lipoprotein(oxLDL) is one of important risk factors on
atheroscle and atherosclerotic diseases.Moreover,Platelet endothelial
cell adhesion molecule (PECAM) may induce rapid adhesion of platelets
to endothelium that is one of the earliest events in atherosclerosis
and inflammation.It was of interest to determine whether oxLDL may
induce PECAM gene expression in endothelial cells.In this
paper,cultured endothelial cells from human umbilical veins were
treated with oxLDL or LDL,and total RNA were iso lated.After
electrophoresis and Northern blot,hybridizations were done with human
PECAM probe and β-actin probe.The results showed that the cells were
grown well in EGMUV media and the cells were died after 24h treated
with oxLDL but not injured after 6h treatment.In LDL treated
group,the cells were less died in 24h.The hybridizative bands were
found with β-actin probe hybridization.However,PECAM gene was not
expressed.The results indicated that oxLDL may injure the endothelial
cells but it can not induce PECAM gene expression of endothelial
cells.
