胰高血糖素抗血清制备与放免测定研究初步报告

摘要 以DEAE-SephadexA-50纯化胰高血糖素,用40%PVP-胰高血糖素作免疫原制备抗血清,其滴度为1∶5500,亲和常数K为1.35×10^(12).LM^(-1);应用WhatmanCF-11纤维素粉纯化标记抗原纯度达92%;采用PEG分离系统建立了简便、灵敏的血浆胰高血糖素放射免疫分析法,其可测范围为10~2000pg/ml,批内和批间变异系数4.4%和5.3%;方法回收率102%。20例正常人空腹血浆胰高血糖素均值107.8pg/ml,50例糖尿病患者组空腹血浆胰高血糖素均值193.8pg/ml。 Pharmaceutical Glucagon(Novo)Was Purified With DEAE-Sephadex A-50.Using 40% PVP-glucagon as the immunoantigen, antiserum was Prepared in Six rabbits. The titer Was 1: 5500 and the affinity constant was 1.85×1012L. M-1. The iodine-abelled antigen was Purified to 92% wth Whatman CF-11 cellulose powder. Applying PEG dissociation system, a convenient Yet Sensitive method of RIA of plasma glucogen was estnblished, with assayable glucagon levels ranging from 10 fo 2000pg/ml and a recovery rate of 102%.The intra-and inter-assay coefficents of variation(cv)were 4.4% and 5.3% respectively. The mean value of fasting plasma glucagon of 30 normal individual was 107.8pg/ml. The mean value of fasting plasma glucagon of 50 patients with diabetes mellitus was 193.8pg/ml.
