流式细胞术检测CO_2气腹对结肠癌细胞表面黏附分子的影响 被引量:15

摘要 目的:探讨不同压强下持续性CO2气腹对结肠癌细胞表面黏附分子表达的影响。方法:建立体外气腹模型,选用人结肠癌细胞株SW1116,分别在不同压强CO2(6mmHg、9mmHg、12mmHg和15mmHg)以及常规条件下暴露1h。应用流式细胞技术(FACScan)检测黏附分子E鄄钙黏着素(E鄄cadherin)、ICAM鄄1、CD44、E鄄选择素(E鄄selectin)于处理后0h、12h、24h、48h及72h在结肠癌细胞表面的表达。结果:SW1116经6mmHg压强CO2气腹处理后,表面ICAM鄄1、CD44表达显著增强,9mmHgCO2气体处理处理后,E鄄选择素、E鄄钙黏着素和CD44表达显著增高,15mmHg压强CO2处理后,E鄄钙黏着素表达增强,与处理前表达水平的差异均有显著性(P<0.05),上述黏附分子的表达在各压强CO2处理后72h之前均降至处理前水平(P>0.05)。或低于处理前水平(P<0.05)。随CO2气腹压强的增高,E鄄钙黏着素、CD44和ICAM鄄1表达量逐渐降低,在处理后0h,差异均达到显著性(P<0.05)。结论:CO2气腹能对肿瘤细胞表面的黏附分子表达产生一过性双向影响。随着CO2气腹压强的增高,可抑制黏附分子表达的增强。 Objective To investigate the influence of CO2-insufflation and its pressure on the expression of a number of adhesion molecules, which were reported to be related to the metastasis of colorectal cancer. Methods With an artificial pneumoperitoneum model in vitro, SW1116 colon carcinoma cells (human) were exposed to CO2-insufflation under different pressure (6 mm Hg, 9 mm Hg, 12 mm Hg and 15 mm Hg) for 1 hour. E-cadherin, ICAM-1, CD44 and E-selectin were measured by flow cytometry 0, 12, 24, 48 and 72 hours after CO2-insufflation. Control groups were exposed to room air. Results The expression of ICAM-1 and CD44 of the SW1116 cell line increased significantly under 6 mmHg CO2-insufflation, under 9 mmHg, E-selectin, E-cadherin and CD44 increased significantly, and when the pressure was 15 mmHg, E-cadherin increased (P<0.05). The expression of these adhesion molecules returned to or even decreased to less than the control level by 72 h(P<0.05). With the ascent of the CO2-insufflation pressure, the increasing expression of the E-cadherin, CD44 and ICAM-1 were restrained in some time points, especially at 0 h after CO2-insufflation (P<0.05). Conclusions The expression of the aforementioned adhesion molecules is effected temporarily in the translation level in both directions after the continuous CO2-insufflation. The pressure of the CO2-insufflation also plays an important role in restraining the expression of the adhesion molecules.
