摘要
目的 通过观察脑源性神经生长因子 (BDNF)对顺铂诱导的神经母细胞瘤 (NB)细胞株生长与凋亡的影响 ,探讨TrKB BDNF通路在NB化疗耐药中的作用。方法 应用四甲基偶氮蓝比色 (MTT)法检测不同浓度BDNF对顺铂诱导人NB细胞株SH SY5Y的生长的影响 ,应用流式细胞仪 (FCM )检测BDNF对顺铂诱导的人NB细胞株SH SY5Y的凋亡率的影响。结果 BDNF可阻断顺铂对TrkB表达的SH SY5Y细胞的杀伤作用 ,且其阻断能力的大小与BDNF浓度有关 ,但不能阻断顺铂对无TrkB表达的SH SY5Y细胞的杀伤作用。顺铂对NB细胞的杀伤作用是通过诱导细胞凋亡 ,BDNF对顺铂这一作用的阻断是通过抑制细胞凋亡而实现的。结论 TrkB BDNF通路在TrkB表达的NB化疗耐药机制中起重要作用。
Objective To explore the role of the TrkB-BDNF pathway in the resistance to chemotherapy in neuroblastoma (NB).Methods The effects of the different concentrations of BDNF on the growth of the human NB cell line SH-SY5Y induced by cisplatin were detected by the method of MTT.FCM was used to detect the effects of the BDNF on the apoptotic rate of the SH-SY5Y induced by cisplatin.Results BDNF could block the role of cisplatin on TrkB-expressing SH-SY5Y in a concentration-dependent manner,but couldn't block the role of cisplatin on SH-SY5Y lack of TrkB.Cisplatin killed the NB cells by inducing apoptosis and BDNF blocked the role of cisplatin by inhibiting apoptosis.Conclusion TrkB-BDNF pathway may play an important role in mediating chemotherapy resistance in NB cells expressing high levels of TrkB.
