脑源性神经生长因子对顺铂诱导的神经母细胞瘤细胞株生长与凋亡的影响 被引量:5

摘要 目的 通过观察脑源性神经生长因子 (BDNF)对顺铂诱导的神经母细胞瘤 (NB)细胞株生长与凋亡的影响 ,探讨TrKB BDNF通路在NB化疗耐药中的作用。方法 应用四甲基偶氮蓝比色 (MTT)法检测不同浓度BDNF对顺铂诱导人NB细胞株SH SY5Y的生长的影响 ,应用流式细胞仪 (FCM )检测BDNF对顺铂诱导的人NB细胞株SH SY5Y的凋亡率的影响。结果 BDNF可阻断顺铂对TrkB表达的SH SY5Y细胞的杀伤作用 ,且其阻断能力的大小与BDNF浓度有关 ,但不能阻断顺铂对无TrkB表达的SH SY5Y细胞的杀伤作用。顺铂对NB细胞的杀伤作用是通过诱导细胞凋亡 ,BDNF对顺铂这一作用的阻断是通过抑制细胞凋亡而实现的。结论 TrkB BDNF通路在TrkB表达的NB化疗耐药机制中起重要作用。 Objective To explore the role of the TrkB-BDNF pathway in the resistance to chemotherapy in neuroblastoma (NB).Methods The effects of the different concentrations of BDNF on the growth of the human NB cell line SH-SY5Y induced by cisplatin were detected by the method of MTT.FCM was used to detect the effects of the BDNF on the apoptotic rate of the SH-SY5Y induced by cisplatin.Results BDNF could block the role of cisplatin on TrkB-expressing SH-SY5Y in a concentration-dependent manner,but couldn't block the role of cisplatin on SH-SY5Y lack of TrkB.Cisplatin killed the NB cells by inducing apoptosis and BDNF blocked the role of cisplatin by inhibiting apoptosis.Conclusion TrkB-BDNF pathway may play an important role in mediating chemotherapy resistance in NB cells expressing high levels of TrkB.
