高效液相色谱法测定朱砂根中岩白菜素含量

Determination of Bergenin in Ardisia Crenata Sims. By HPLC

目的 :为控制朱砂根药材质量 ,建立测定朱砂根中岩白菜素含量的高效液相色谱方法。方法 :采用HypersilODS色谱柱 (5 μm ,2 0 0mm× 4 6mmi.d .,Agilent) ;柱温 2 5℃ ;流动相为甲醇—水 (体积比为 2 4∶76 ) ;流速为 1 0ml.min- 1 ;检测波长为 2 74nm。结果 :岩白菜素的进样量为 1 0 0～ 12 0 1μg时与峰面积呈良好线性关系 ,平均回收率 99 6 6 % ,RSD为 1 0 1% (n =6 ) ;结论 :本法准确、灵敏度高、重现性好 ,可作为朱砂根药材质量控制的含量检测方法。

Objective: To control the quality of Ardisia crenata Sims., The contents of bergenin was determined by HPLC . Method: The HPLC system consisted of Hypersil ODS (200mm×4.6mm i.d., 5μm); column temperature 25℃; mobile phase of methanol-water (24:76 in volume ratio), flow rate 1.0ml/min; detecting wavelength 274nm.Result: The linear rages of determination of bergenin was 1.00～12.01μg, the mean recovery rate is 99.66%, RSD=1.01%. Conclusion: The method is accurate, sensitive and with good repeatability, and can precisely determine bergenin content in Ardisia crenata Sims.
