摘要
目的 :研究脂质体混悬液对荧光素钠在大鼠皮肤分布的影响。方法 :以0 125 %荧光素钠溶液为对照,在激光共聚焦显微镜下观察不同时间点0 125 %荧光素钠脂质体混悬液 (试验组 )荧光素钠在皮肤表皮、真皮分布的情况。结果 :试验组表皮及真皮荧光强度峰值与对照组比较均具有统计学差异 (P<0 05) ;试验组表皮及真皮内荧光强度值曲线下面积分别是对照组的2 09倍和2 4倍 ;试验组切片中毛囊周围的荧光强度值大于对照组。结论 :脂质体混悬液能使药物在皮肤组织中保持较高的浓度 ,从而提高其生物利用度 ;毛囊是脂质体制剂中药物扩散的重要途径。
OBJECTIVE:To study effect of liposome suspension on distribution of fluorescein sodium in the rat skin.METHODS:Using0.125%fluorescein sodium(NaFl)solution as control,the distribution of NaFl in epidermis and dermis at different time periods was observed under confocal laser scanning microscope in0.125%NaFl liposome suspension group(trial group).RESULTS:The fluorescent intensity peaks in epidermis and dermis of trial group were higher than those in skin layers of control group(P<0.05).The AUCs of fluorescent intensity in both epidermis and dermis in trial group were2.09~fold,2.4~fold that of control group respectively.The fluorescent intensity of NaFl around hair follicular structures in the skin was much higher in trial group than in control group.CONCLUSION:Liposome suspension can deposit drug in the skin and keep high drug concentration in skin,increase topical drug bioavailability.Hair follicular structures is an important pathway in skin penetration.
