摘要
目的 检测痢疾志贺氏I型菌特异性多糖 (O SP)和破伤风类毒素结合物O SP TT的生物学活性。方法 应用ELISA间接法测定O SP、O SP TT分别免疫小鼠后其血清中IgG、IgM的水平 ;应用补体介导的杀菌活性实验检测血清的体外杀菌活性。结果 单独使用O SP免疫小鼠后抗体无显著性升高 (P >0 0 5 )。O SP TT免疫注射后三次之间IgG和抗TT抗体均有显著性增高 (P <0 0 1 )。多糖 蛋白结合物免疫后的血清中抗体可激活补体产生杀菌作用的效价为 1∶1 60。结论 多糖 蛋白结合物注射后对O SP和TT均可加强免疫应答 。
Objective To investgate biological activity of conjugate composed of the O-specific Polysaccharide of Shigella dysenteriae type I bound to tetanus toxoid. Methods Serum O-SP and TT antibodies were measured by enzyme linked immuno sorbent assaying . The bactericidal activity of serum O-SP antibody was measured by complement dependent bactericidal experiments.Results The result shows alone the O-SP of Shigella dysenteriae of type I was not elicited detectable serum antibodies( P>0.05). A statistically significant rise in both IgG and IgM antibodies could be elicited by the O-SP-TT conjugate immunized mice after each injection(P<0.01). The resultant O-SP-TT conjugate can elicit high levels of IgG O-SP antibodies and booster responses in young mice after the second injection. TT-Ab were elicited by conjugate after the second and third injections.Conclusion The booster responses occur after the second and third injection of the O-SP-TT conjugate as a results of the immunological memory response. The O-SP antibodies in serum elicited by O-SP-TT conjugate had strongly bactericidal activity in confirmation with complement dependent bactericidal text in this study.
