摘要
本文研究了Pre-S2蛋白体液免疫反应的精细特异性。结果表明小鼠对Pre-S2蛋白的抗体反应部位主要局限于14—24共11个连续的氨基酸残基内,与我们理论预测的B细胞表位相吻合;人的抗Pre-S2阳性血清除识别与小鼠抗血清及其Pre-S2特异性单抗同一肽段外,还识别1—13肽段。这些结果对于设计新一代合成疫苗及诊断试剂有意义。
The fine specificity of the humoral immune response to the Pre-S2 region of the Hepatitis B surface antigen (HBsAg) was studied. It was demonstrated that the murine antibody response to the Pre-S2 region is dominantly focused on residues 14 though 24. In quantitative antibody inhibition studies it was demonstrated that all the Pre-S2 specific monoclonal antibodies employed in the study except McAbl20-603 were capable of inhibition each other. Finally, human HBV-infected patient sera were shown to possess anti-pre-S2 region antibodies that recognize sequences largely common with the murine sera. These results have implications relavant to the design of synthetic and recombinant second generation HBV vaccines and diagnostic reagents.
