Spontaneous Secretion of Interleukin-6 (IL-6) by Primarily Cultured Astrocytes of New-born Mice

摘要 本文选用出生3天以内的小白鼠,以酶消化方法制备乳鼠脑星形胶质细胞进行原代培养,按不同时相收获上清,并对其上清进行白细胞介素6(IL—6)生物活性检测。结果表明各组上清中均显示6明显的IL—6活性,在一定范围内呈剂量依赖关系。不同浓度的乳鼠脑星形胶质细胞培养上清支持IL—6依赖细胞株MH60.BSF_2细胞的体外培养,细胞生长良好,并均有增殖。提示:原代培养的脑星形胶质细胞在无任何刺激情况下可自发分泌IL-6。 The astrocyte from new-born balb/c mice (less than 3 days) was separated by trypsin (0. 25%) enzymatic method and cultured for different time. The supernatant of the cell culture was assayed for IL-6 with MH60. BSF-2 cell line, the results showed that the supernatant from the cells culture (2×1O4/ml) of day 1 to day 3 (dl-d3), d3 -d5and d5 -d14 contained high titer of IL-6 (>123. 20u/ml, 46. 66 u/ml. and 75. 90u/ml respectively). In another experiment the supernatant of cell culture(1×105/ml) of dl - d6 and d6 - dll contained IL-6 of 25. Ou/ml and 12. 5u/ml. The dose-dependent proliferation of MH60. BSF-2 on supernatant of astrocyte culture was observed. It is indicated that the astrocytes of new born mice could secret high level of IL-6 spontaneously, and the shorter the astrocytes leaves from the physiological environment (eg. brain), the more the IL-6 is secreted, suggesting that the IL-6 as a new neurotrophic factor, plays an important role in differentiation of nerve cells. The discovery of the new function of IL-6 on nerve cells and new producer (astrocytes) enriched the theory of neuroendocrineimmune system.
