环缩酚酞抑制血管内皮细胞生长因子诱导的血视网膜屏障破坏 被引量:1

Hep-A and Hep-B reduced vascular endothelial growth factor induced breakdown of blood-retinal barrier in mice

摘要 目的 观察两种新的环缩酚酞 (Hep)衍生物 Hep- A、Hep- B对血管内皮细胞生长因子(VEGF)诱导的血视网膜屏障破坏的效应。 方法 将 C5 7BL / 6 J小鼠分为正常对照组、阳性对照组、Hep- A、Hep- B治疗组 ,对照组皮下注射安慰剂 ,治疗组分别皮下注射 Hep- A、Hep- B 10 mg/ kg,2次 / d,持续 5 d。 5 d后 ,阳性对照组、Hep- A、Hep- B治疗组玻璃体内注射 1μl 10 - 6 mol/ L重组人类 VEGF诱导血视网膜屏障破坏 ,6 h后腹腔内注射 3.7× 10 4 Bq/ g的 3H-甘露醇。处死小鼠并测量视网膜、肺、肾组织的放射活性 ,分别计算各组的视网膜与肺、视网膜与肾放射性比率 ,用方差分析比较各组间差异。 结果 正常对照组的视网膜与肺和视网膜与肾放射性比率分别为 0 .38± 0 .0 4和 0 .2 1± 0 .0 3;阳性对照组为 1.0 5±0 .11和 0 .4 6± 0 .0 4 ,均较正常对照组显著升高 (P值均 <0 .0 1) ;Hep- A治疗组分别为 0 .5 9± 0 .0 6和0 .32± 0 .0 3,均较阳性对照组显著降低 (P值均 <0 .0 1) ;Hep- B治疗组分别为 0 .5 4± 0 .0 4和 0 .35± 0 .0 3,均较阳性对照组显著降低 (P值分别 =0 .0 1和 <0 .0 1)。 结论 Hep- A和 Hep- B可以抑制 Objective To investigate the effects of Hep-A and He p-B on vascular endothelial growth factor (VEGF)-induced breakdown of blood-r etinal barrier. Methods The mice were subcutaneously injected vehicle, Hep-A or Hep-B 10 mg/kg twice a day for 5 days. Then, 1 μl of 10 -6 mol/L VEGF were intravitreous injected. After 6 hours, 13.7×10 4 Bq /g 3H-mannital were injected intraperitoneally. The mice were sacrificed and the retinas, lungs, kidneys were removed and examined for radioactivity. The result were analyzed using SPSS software to calculate and compare retina/lung and et ina/kidney leakage ratio among groups of different treatment. Result The retina/lung and retina/kidney leakage ratio were 0.38±0.04 an d 0.21±0.03 respectively in normal mice; increased significantly to 1.05±0.11 and 0.46±0.04 respectively in model mice, both P<0.01 compared to those in normal mice; decreased to 0.59±0.06 and 0.32±0.03 respectively in mice treated with Hep-A, both P<0.01 compared to those in model mice; decreased 0.5 4±0 .04 and 0.35±0.03 in mice treated with Hep-B, both P<0.01 compared to thos e in model mice. Conclusion Hep-A and Hep-B can significantl y reduce VEGF-induced breakdown of blood-retinal barrier in mice.
