芒果甙对白血病K562细胞增殖抑制作用的研究 被引量:19

摘要 目的 :研究芒果甙在体外对慢粒白血病细胞系 K5 6 2细胞生长的抑制作用。方法 :采用 MTT法、集落形成试验及生长曲线绘制分别观察不同浓度芒果甙对 K 5 6 2细胞生长的影响。结果 :3种方法均显示不同浓度的芒果甙在不同时间对 K5 6 2细胞均有抑制作用 ,并随浓度升高和作用时间延长而抑制作用增强 ,其中 MTT法显示 ,2 5、5 0、1 0 0、1 5 0、2 0 0μmol/ L浓度芒果甙作用 K5 6 2细胞 72 h后 ,其抑制率分别为 31 .99%、5 0 .74 %、6 0 .2 4 %、6 2 .6 5 %和 70 .4 1 % ;2 0 0μmol/ L浓度芒果甙作用于K5 6 2细胞 2 4、4 8、72和 96 h后 ,其抑制率分别为 4 8.1 9%、6 8.88%、74 .38%、76 .93%和 83.5 0 %。结论 :芒果甙能明显抑制K5 6 2细胞的增殖 。 Objective:To study the effects of growth inhibition of mangiferin on K562 leukemia cells in vitro.Methods:The effect of different concentrations of mangiferin on the proliferation of K562 leukemia cells were examined by MTT assay, colony formation assays and growth curves.Result:All of the 3 methods showed that different concentrations of mangiferin can inhibit the K562 cells proliferation in different times and the inhibition effect were dependent on time and concentration;and the MTT assays showed that the inhibition of K562 cells was 31 99%,50 74%,60 24%,62 65% and 70 41% respectively with (25~200 μmol/L) mangiferin treatment for 72 hours , the inhibition of K562 cells was 48 19%,68 88%,74 38%,76 93% and 83 50% respectively with 200μmol/L mangiferin treatment for 24,48,72 and 96 h Conclusion:The proliferation of K562 cells was markedly inhibited by mangiferin in a dose-dependent and time-dependent manner.
