日本血吸虫RhoGTPaseDNA及重组蛋白免疫保护效果研究 被引量:4

Protective Immunity Studies of Sj-Rho GTPase Dnavaccine and Recombinant Vaccine Against Schistosoma Japonicum

摘要 目的 研究日本血吸虫中国大陆株Rho家族小分子G蛋白(Sj-Rho GTPase)DNA疫苗及重组蛋白疫苗免疫保护效果。方法 将pcDNA3.1-SjRho GTPase DNA疫苗和pGEX-5X-3原核载体所表达的重组蛋白疫苗三次分别或联合免疫昆明鼠,3周后,每鼠感染40条尾蚴,42 d后剖杀小鼠,记数成虫和肝脏虫卵数。结果 与佐剂或空白质粒对照组相比,DNA疫苗、重组蛋白疫苗及联合免疫分别获得22.16%(P<0.01)、18.71%(P<0.05)和39.20%(P<0.001)的减虫率以及34.55%、20.07%和49.55%(P<0.001)的肝组织减卵率。结论Sj-Rho GTPase DNA疫苗及重组蛋白疫苗均可诱导小鼠产生一定的免疫保护作用,且联合免疫方案保护效果好于单一疫苗。 Objective To study the protective immunity induced by the DNA vaccination and recombinant vaccination of Sj - Rho GTPase of Schistosoma japonicum (Sj). Methods All mice were immunized three times with naked plasimids pcDNA3.1 - SjRho GTPase, protein of Sj - Rho GTPase or both the DNA and protein boosting vaccination in an interval of 3 weeks, and challenged each with 40 cercariae of S.japonicum at 3 weeks after final immunization. At the 42nd day after challenge, all mice were sacrificed,and the numbers of worms and eggs in the livers of mice were counted. Results Compared with the control group, the worm reductions of the DNA vacci natin and the recombinant protein vaccination and the DNA priming - protein boosting vaccination were 22.16% (P <0.01), 18.71%(P <0.05) and 39.20% (P < 0.001) respectively; the egg reductions were 34.55%,20. 07% and 49.55%( P < 0.001) respectively. Conclusion DNA vaccination and recombinant GTPase could induce partial protective immunity against S.japonicum in mice. The DNA priming-protein boosting vaccination regimen was better than the single vaccination.
