硝酸甘油预处理对缺血再灌注心肌中性粒细胞浸润的影响

摘要 目的 初步探讨硝酸甘油预处理是否影响缺血再灌注心肌中性粒细胞(PMN)的浸润。方法 SD大鼠30只随机分为3组,在在体大鼠心肌缺血60 min再灌注4 h模型上分设:假手术组、生理盐水对照组、硝酸甘油组。用组织切片方法及髓过氧化物酶法(MPO)检测PMN浸润情况;用Evans Blue加TTC染色法测梗塞范围,并测血清CK活性。结果 与生理盐水组相比,硝酸甘油组大鼠心肌梗塞范围明显减小,血清CK活性明显降低,MPO活性降低,PMN浸润明显减轻。结论 硝酸甘油(NTG),早期预处理(PC)可减轻大鼠在体缺血再灌注心肌PMN的浸润。 Objective To investigate the effects of NTG - PC on the neutrophil infiltration in a murine ischemia/ reperfusion model. Methods Adult SD rats were randomly divided into 3 groups: sham operation group, saline control group, NTG group (NTG pretreatment 1 h before I/R). All groups, except sham grouop, were subjected to 60 min of occlusion and 4 h of reperfusion. Neutrophil infiltration in focal myocardial tissue after I/R was determined. Infarct size(IS), as a percentage of the area at risk (AAR), was determined by Evan's blue TTC staining. Serum level of CK activity and myeloperoxidase activity in cardiac tissue were also been determined. Results I/R resulted in a significant myocardial injury and marked PMN accumulation. Administration of NTG 1 h before I/R significantly inhibited the myocardial inflammatory response as evidenced by reduced PMN accumulation (MPO activity), and markedly attenuated myocardial infarct size and content of serum CK. Conclusion NTG PC inhibits PMN accumulation in I/R myocardium in vivo, thus, provides myocardial protection.
