隔药灸对溃疡性结肠炎大鼠结肠IL-1β、IGF-1表达影响的研究

Study on the Expressions of IL-1β and IGF-1 in Ulcerative Colitis Rats Treated by Herbs-partition Moxibustion

目的应用荧光定量PCR观察隔药灸对溃疡性结肠炎大鼠结肠IL1β、IGF1表达的影响。方法将溃疡性结肠炎大鼠随机分为模型组和隔药灸组,另设正常对照组。隔药灸组穴位选取“天枢”,采用隔药灸进行治疗,并应用实时荧光定量PCR方法检测IL1βmRNA、IGF1mRNA的表达。结果溃疡性结肠炎大鼠结肠IL1βmRNA、IGF1mRNA表达异常升高,隔药灸治疗能降低其表达。结论隔药灸可通过调节IL1β、IGF1基因的表达,消除UC大鼠肠道炎症,并可能起到防治肠纤维化的作用。

Objective: To observe the influence of herbs-partition on interleukin-1beta messenger RNA (IL-1βmRNA), Insulin-like growth factor-1 messenger RNA (IGF-1 mRNA) expressions in ulcerative colitis(UC) rats using fluorescence quantitative polymerase chain reaction.Methods: The rats were randomly divided into 3 groups as normal control group, UC model control group and herbs-partition moxibustion group. The rats in herbs-partition moxibustion group were treated by herbs-partition moxibustion on Tianshu(ST 25, bilateral). Fluorescence quantitative polymerase chain reaction was employed to detect IL-1βmRNA and IGF-1 mRNA expressions.Result: The expression of IL-1βmRNA and IGF-1mRNA in UC rats abnormally increased, and reduced after treating by herbs-partition moxibustion.Conclusion: By adjusting the expression of many genes such as IL-1β, IGF-1mRNA, herbs-partition moxibustion may eliminate the intestinal inflammation in UC rats and maybe prevent the happening from intestinal fibrosis change.
